AplaGen announces publication of a patent application for a novel technology that increases half-life of proteins in vivo

2009/12/07 - AplaGen GmbH announced today the publication of a patent application describing an innovative formulation allowing elongation of e.g. the half-life of recombinant factor VIII used in hemophilia. This innovative method increases the half-life of complex and/or multifunctional proteins in human and animal circulation, can reduce infusion rates and thereby increase patients quality of life. Aside from the increased quality of life resulting from fewer treatments, it also reduces the cost of the treatment.

Additionally, this non-covalent technology provides the advantage that proteins used in therapy do not have to be chemically modified in order to prolong their half-life. Instead the conjugates are incorporated into the existing pharmaceutical composition in order to exert their function. This also creates a regulatory benefit because there is no change to the active pharmaceutical ingredient (API), only the formulation of the product.

AplaGen presents its major product SESTide at the 2009 Luebeck Conference on EPO as an efficient EPO mimetic substance which, apart from being an agent applicable for fighting anemia, can also be used as an antagonist for (endogenous) EPO in oncological indications

2009/08/01 - At the 8th International Luebeck Conference on EPO, AplaGen reported on the present state of its major pharmacological project "SESTide" (supravalent erythropoiesis stimulating peptide). It was shown that SESTide, being a dimeric EPO mimetic peptide (EMP) conjugated to hydroxyethyl starch (HES), shows very competitive efficacy as an erythropoiesis stimulating agent (ESA) both in various in vitro assays and in animal (pharmacodynamic and pharmacokinetic) experiments. Nevertheless, and most remarkably, SESTide and its prototype substance AGEM400(HES) were shown to have a different activity spectrum than recombinant EPO. Whereas AplaGen’s EMP behaves as an effective ESA in in vitro and in in vivo models of erythropoiesis, it was shown to have very poor to absent activity in three leukemic cell lines which respond, by prolonged survival, to recombinant ESA. In one leukemia cell line, UT7, in which the EMP did not show activity at all, the peptide antagonized the survival-promoting effect of EPO by competing for binding to the EPO receptor. In preparation of further studies on the differential effects of recombinant ESA and EMP in other models of malignancies, AplaGen has already filed a patent application on the use of EMP as an agent to be used to antagonize the potentially harmful effects of EPO (endogenous or externally administered recombinant ESA) on malignancies expected to be stimulated by this cytokine (EPO-responsive malignancies).

AplaGen closes financing round

2007/06/29 - The biopharmaceutical company AplaGen GmbH in Baeweiler near Aachen in Germany aquired 3,5 Mio. Euro venture capital for further preclinical development of its peptide products. Investors in this financing round were the present shareholders, lead by Intelligent Venture Capital, Aachen. Further investors are NRW.Bank Venture Fonds, Düsseldorf and as new shareholders KfW and some private investors. In addition to the equity contribution in this financing round the company was able to win significant research grants.

The capital allocation will finance the further preclinical development of AplaGen´s lead product Hemomer®. The patented drug is a fully synthetic EPO mimetic peptide stimulating hematopoiesis in anemia patients. Recombinant drugs already address a 12 b$ market and are prescribed in anemia cases caused by cancer, renal dysfunction or chronic kidney disease. In comparison with the existing recombinant EPO´s Hemomer® offers several advantages for the patients and can be produced very cost efficient.

Apart from HemoMer® AplaGen develops with Thrombomer® a closely related Cytokine mimetic peptide. This thrombopoietin mimetic peptide is a stimulator of blood platelet formation in the bone marrow and therefore responsible for bleeding control and coagulation.
